The Holtek Data sheet for the HT18K33 is a fairly decent 47 page description of the part you have selected. I found no big mystery within the data sheet and it seems completely straightforward to setup and use the device.

Holtek has chosen a somewhat unfortunate documentation to specify the bits of command bytes sent to the part as D15-D14-D13-D12-D11-D10-D9-D8. These are in no way really meant to specify that any real 16 bit data is being sent to the part. So you should just consider the commands to be a byte that gets sent from the MCU over the I2C bus. At least they use this D15->D8 terminology consistently throughout the data sheet.

Note that I have never used this IC chip and so cannot offer any specific detailed advice over what I can read in the data sheet. I presume you have the data sheet and are able to read it and understand how the part works inside. If you have specific questions or doubts about any part of the data sheet then certainly come back here with your concerns and we'll see if we can help to interpret it for you.
