The Holtek Data sheet for the HT18K33 is a fairly decent 47 page description of the part you have selected. I found no big mystery within the data sheet and it seems completely straightforward to setup and use the device.
Holtek has chosen a somewhat unfortunate documentation to specify the bits of command bytes sent to the part as D15-D14-D13-D12-D11-D10-D9-D8. These are in no way really meant to specify that any real 16 bit data is being sent to the part. So you should just consider the commands to be a byte that gets sent from the MCU over the I2C bus. At least they use this D15->D8 terminology consistently throughout the data sheet. Note that I have never used this IC chip and so cannot offer any specific detailed advice over what I can read in the data sheet. I presume you have the data sheet and are able to read it and understand how the part works inside.
